Its effects on breast cancer cell proliferation and invasiveness were also examined. The effect of C/EBPβ on breast cancer cell cycle was further analyzed using flow cytometry and immunoblotting. The effect of C/EBPβ knockdown on colony formation in soft agar was also analyzed. Our results demonstrated that C/EBPβ was upregulated in human breast cancer cells. Downregulation of C/EBPβ by transfection of C/EBPβ-specific small hairpin RNA (shRNA) suppressed the viability and proliferation of breast cancer cells. Also, knockdown of C/EBPβ induced G0/G1 arrest in breast cancer cells. Furthermore, knockdown of C/EBPβ significantly suppressed the migration and invasiveness of breast cancer cells.
